Usually, it depended on the job. But normally, two brakemen, one called the head end brakeman, riding on the locomotive or tender in the dog house, the other on the caboose, and a conductor.

On jobs that had a lot of switching to do, like a local freight, there was a third brakeman. So either 5 or six men depending on the circumstance.
